Protection conferred by a live Salmonella Enteritidis vaccine against S. Enteritidis during the period of pullet rearing

Four trials were carried out in laying pullets to evaluate the protection of a live attenuated Salmonella Enteritidis vaccine, Tad Salmonella Vac E® (Lohmann Animal Health, Germany) against S. Enteritidis challenge.

Methodology

 

Chickens: Salmonella free 1 day-old commercial layer female chicks. (Lohmann Brown)

Vaccination route: By gavage into the crop.

Vaccination dose: 1 to 5 x 108 Colony Forming Units per dose.

Culture media: Brilliant Green plus Nalidixic acid agar and Tetrationate broth.

Faecal samplings: 5 times by cloacal swabbing.

Organs cultured: Livers, ovaries, spleens and caeca.

Experimental design

 

Trials 1, 2 and 3: 42 vaccinated chickens and 42 unvaccinated ones.

Trial 4: 68 chickens were divided into 4 experimental groups of 17 chickens each

Group 1: Vaccinated at the 1st day of life

Group 2: Vaccinated at the 7th week of life

Group 3: Vaccinated at the 1st day and 7th
week of life

Group 4: Control unvaccinated.

Challenges Dose: 2 - 5 x 109 Colony- Forming Units per dose.

Strain: Virulent S. Enteritidis 285/94 Nalidixic acid resistant.

Trial 1: 6th week of life.

Trial 2: 8th week of life.

Trial 3: 12th week of life.

Trial 4: 16th week of life.

At the 5th day post challenge: 4 - 5 chickens per group were sacrificed.

At the 15th day post challenge: All remaining chickens were sacrificed.

Results

 

  • Fewer vaccinated chickens excreted S. Enteritidis in comparison to unvaccinated ones.

 

  • Fewer organs were found to be positive for the presence of S. Enteritidis.
     

Conclusions

 

  • Administration of one oral dose of Tad Salmonella Vac E® at the 1st day of life protects the chicken at least until the 8th week of life.

 

  • Two oral doses at the 1st day and 7th week of life of Tad Salmonella Vac E® during the period of pullet rearing are recommended to afford protection against S. Enteritidis infection during the rearing period.
